Introduction
Chloramphenicol (CAP) is a broad spectrum antibiotic. It is commonly used for treating many kinds of disease of domestic fowl because of its efficiency and low price. However, using chloramphenicol will inhibit the hematogenesis function of bone marrow, moreover, it causes induces aplastic anemia and agranulocytosis. Recently, the Chloramphenicol residues in food threaten the health of human has been severely.
The Chloramphenicol chromatographic diagnostic kit is based on the specific immuno-chemical reactions between antigen and antibody. It relies on the competition between Chloramphenicol residues in the sample and? the Chloramphenicol immobilized on T line on the membrane for the Chloramphenicol antibody-dye conjugate. Since the drug has been presenting in the sample, it will compete with the drug immobilized on T line, to the limited amount of dye-antibody. As a sufficient amount of drug in the sample is presenting, the drug will saturate the antibody. Consequently, it will show an extremely light or even invisible T line, indicating a positive result. On the other hand, if there is a negative sample (or the amount of the drug is lower than the minimum detectable concentration, 0.3 ppb), it will generate two obvious lines in both the T and C line section.

Operation Procedure
A. Precaution
1、 Open the package just before use to avoid the test cassette being damped.
2、 Avoid touching the membrane in the cassette.
3、 Every test cassette and dropper is single use only. Please do not reuse.
4、 Ethyl Acetate and n-Hexane are highly flammable, please keep away from fire.
5、 Please use 50 mL conical tube made of polypropylene to prevent corruption during extraction procedures.
6、 Store at 8~30 ℃. Do not freeze.
B. Sample preparation
Please read Precaution before use.
1、 Weigh 5±0.05 g well homogeneous sample in a 50 mL tube, and add 5 mL distilled water. shake properly.
2、 Add 10mL Ethyl Acetate and vortex for 5 min. Centrifuge at 4000 rpm for 10 min at room temperature.
3、 Take 8 mL of upper phase to a clean glass tube and Then blow to dry completely with the nitrogen at 60 ℃ waterbath.
4、 Dissolve the residue in 2 mL n-hexane thoroughly and then add 0.5 mL distilled water, vortex for 30 seconds.
5、 Centrifuge at 4000 rpm for 10 min at room temperature.
6、 Discard the upper phase and take80 µL of the bottom layer solution for assay.
C. Operation procedures
1、 Open a package and take out the test cassette.
2、 Use the provided dropper to obtain the sample. Drop 3 drops (about 120 μL) in the Sample collect region.
Note: Do not drop the sample directly to the test result region. It will affect the result.
3、 Wait for 5-10 min to read the result.
Note: Do not wait more than 15 min to read the result. Wrong Interpretation will be caused.
D. Test Result Interpretation
1、 Negative (CAP < 0.3 ppb): T line is even or darker than C line.
2、 Positive (CAP > 0.3 ppb): T line is invisible or lighter than C line.
3、 Invalid test: If C line does not show which means the test cassette is out of efficacy, out of date or improper operation. Please run the test again using another package. If the invalid tests keep happening, please contact the local distributor.
