1. Introduction

Ractopamine (Rac) is one kind of beta-agonist, normally, dissolves in hydrochloride as Ractopamine hydrochloride called Paylean. Rac is used as feed additives for transfering adipose tissue to muscle tissues in pig. Because lots of countries (ex. EU) regard it as growth hormone, import of Ractopamine remaining meat is banned. Consequently, a convenient rapid Ractopamine diagnostic kit is developed and fit in need.

2. Principle

The Ractopamine chromatographic diagnostic kit is based on the specific immuno-chemical reactions between antigen and antibody. It relies on the competition between Ractopamine residues in the sample and? the Ractopamine immobilized on T line on the membrane for the Ractopamine antibody-dye conjugate. If the drug is presenting in the sample, it will compete with the drug immobilized on T line, to the limited amount of dye-antibody. When a sufficient amount of drug in the sample is presenting, the drug will saturate the antibody. Thus it will show an extremely light or even invisible T line, indicating a positive result. If there is a negative sample (or the amount of the drug is lower than the minimum detectable concentration 5ppb), it will generate two obvious lines in both the T and C line section.

4. Precaution

1  Open the package upon request to avoid the test cassette being damped.

2  Use one cassette and one dropper for one test. Store the rest of the test cassette in dark at 4-30 ℃ environment and avoid being damped.

3  Avoid touching the membrane in the cassette.

4  Each package containing one test cassette and one drop is for one time use only. Please do not reuse.

5. Operation procedures

1  Open the aluminum package. Then take out the test cassette.

2  Use the provided dropper to obtain the sample. Drop 3 drops (about 80 μL) in the sample collect region.
Note: do not drop the sample directly in the test result region. It will affect the result.

3  Read the results in 5-30 min.
Note: do not wait over than 30 minutes to read the result. Time will affect the result.

6. Test Result Interpretation

1  Negative (Rac < 5 ppb): T line is even or darker then the C line.

2  Positive (Rac > 5 ppb): T line is invisible and only C line is visible.

3  Invalid test: If C line does not show which means the test cassette is out of efficacy, out of date or improper operation. Please run the test again using another package. If the invalid tests keep happening, please contact the local distributor.
